Lysergicacid2-butylamide(2-Butyllysergamide,LSB)isananalogueoriginallydevelopedbyRichardPiochatElyLilliinthe1950s,butmostlypublicisedthroughresearchconductedbytheteamledbyDavidE.NicholsatPurdueUniversityItisastructuralisomerwiththetwoethylgroupsontheamidenitrogenhavingbeenreplacedbyasinglebutylgroup,joinedatthe2-position.Itisoneofthefewlysegamidederivativestoexceedthepotencyinanimaldrugdiscriminationassays,withthe(R)isomerhavinganED50of33nmol/kgforproducingdrug-appropriateresponding,vs48nmol/kg.Thecorresponding(R)-2-pentylanaloguehashigheraffinityforthe5-HT1Aand5-HT2Areceptors,butislesspotentinproducingdrug-appropriateresponding,suggestingthatthebutylcompoundhasahigherefficacyatthereceptortarget.
